  {"id":24628,"date":"2023-10-05T14:11:55","date_gmt":"2023-10-05T19:11:55","guid":{"rendered":"https:\/\/uwm.edu\/letters-science\/?p=24628"},"modified":"2023-12-04T13:29:35","modified_gmt":"2023-12-04T19:29:35","slug":"uwm-welcomes-a-new-shaw-scientist-with-expertise-on-parkinsons","status":"publish","type":"post","link":"https:\/\/uwm.edu\/letters-science\/in-focus-2023\/uwm-welcomes-a-new-shaw-scientist-with-expertise-on-parkinsons\/","title":{"rendered":"51ÁÔÆæ welcomes a new Shaw Scientist with expertise on Parkinson\u2019s"},"content":{"rendered":"\n<p>With support from the Greater Milwaukee Foundation, An-Phu Tran Nguyen has begun his appointment at 51ÁÔÆæilwaukee as both an assistant professor of biological sciences and as a Shaw Scientist.<\/p>\n\n\n\n<p>Shaw Scientist awards were developed by the Greater Milwaukee Foundation, working with a panel of leading researchers and scientists, to fulfill the wish of Dorothy Shaw.<\/p>\n\n\n\n<p>Shaw, a philanthropist who lost two sisters to cancer, left the bulk of her estate, a $4.2 million gift, to the Foundation with this challenge: Find a way to support emerging scientists at the University of Wisconsin-Madison and the University of Wisconsin-Milwaukee engaged in research in the fields of biochemistry, biological science or cancer research. Each year, scientists from 51ÁÔÆæ or 51ÁÔÆæadison who are in the early stages of their career can be nominated for this award, which provides $200,000 in unrestricted funding.<\/p>\n\n\n\n<p>Tran Nguyen now joins the ranks of those awardees. Tran Nguyen said he is honored to be chosen as a Shaw Scientist and excited to make his own mark at 51ÁÔÆæ. His nascent research program will be supported by the Greater Milwaukee Foundation\u2019s Shaw Fund.<\/p>\n\n\n\n<p>\u201cWhen I started to look up the information about the Greater Milwaukee Foundation, I saw a lot of fantastic scientists who have been awarded over the years with the Shaw Scientist Awards, and I am in admiration of their work. I feel really honored to be funded by the same institution,\u201d Tran Nguyen said. \u201cI cannot say thanks enough for their support.\u201d<\/p>\n\n\n\n<p>Here\u2019s a closer look at one of the newest Shaw Scientists.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">A world traveler<\/h3>\n\n\n\n<p>Tran Nguyen is a native of Vietnam, and he\u2019s keenly aware of the aftereffects of the Vietnam war.<\/p>\n\n\n\n<p>\u201cI\u2019m a lucky generation. We grew up without a war to worry about,\u201d he reflected. \u201cThere was a long isolation of the country for nearly two decades.\u201d<\/p>\n\n\n\n<p>But in the last three decades, the country has been \u201copening up and quickly growing,\u201d allowing Tran Nguyen and his peers to understand just what they were missing.<\/p>\n\n\n\n<p>\u201cThere was a lot to catch up on. We had the idea that we were so behind in technology and education,\u201d he said. \u201cEveryone wanted to try to get out and see the world.\u201d<\/p>\n\n\n\n<p>And see the world he did. Tran Nguyen threw himself into his education, starting with learning French. That made it easy for him to study at Le Mans Universit\u00e9 in France before transferring to the Universit\u00e9 d\u2019Angers, where he earned both his bachelor\u2019s and master\u2019s degrees and specialized in cellular and molecular signaling in health and diseases. He continued his studies with a PhD that he earned at the University of Tuebingen in Germany, funded by the&nbsp;<a href=\"https:\/\/marie-sklodowska-curie-actions.ec.europa.eu\/\" rel=\"noreferrer noopener\" target=\"_blank\">Marie Curie Actions<\/a>&nbsp;program, and then a post-doc position at the&nbsp;<a href=\"https:\/\/www.vai.org\/\" rel=\"noreferrer noopener\" target=\"_blank\">Van Andel Institute<\/a>&nbsp;in Michigan, where he was also promoted to a reasearch scientist position.<\/p>\n\n\n\n<p>Now, he\u2019s settling into his new role as an assistant professor at UW-Milwaukee.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Parkinson\u2019s and genetics<\/h3>\n\n\n\n<p>Tran Nguyen\u2019s research focuses on Parkinson\u2019s Disease (PD) \u2013 specifically, its genetic components.<\/p>\n\n\n\n<p>PD is a neurogenerative disorder. Public figures like Michael J. Fox, Muhammed Ali, Pope John Paul II, and others were diagnosed with Parkinson\u2019s. Around 90,000 Americans are diagnosed with PD each year, according to the Parkinson\u2019s Foundation. Its symptoms include unintended or uncontrollable movements, muscle stiffness, and difficulty with balance and coordination.<\/p>\n\n\n\n<p>\u201cThe main cause of those is a lack of dopamine that is released by dopaminergic neurons in the substantia nigra, a structure located in the basal ganglia. It\u2019s a small part of the brain, but it\u2019s important. Dopamine is a neurotransmitter, a kind 1of a master regulator of mood and movement,\u201d Tran Nguyen explained. \u201cIn PD patients, the dopaminergic neurons are dying, causing a dopamine deficiency.\u201d<\/p>\n\n\n\n<p>But while scientists understand that a lack of dopamine is the culprit of the symptoms, \u201cwe still don\u2019t know what is causing the loss of nigral dopaminergic neurons and not others in PD,\u201d he said.<\/p>\n\n\n\n<p>As more and more scientists have begun studying PD and other neurodegenerative diseases, they have found some genetic links that may point to causes of PD, and potential therapeutic targets. Tran Nguyen has been studying a gene known as LRRK2.<\/p>\n\n\n\n<p>\u201cThe gene is encoding for a protein with the same name: Leucine-rich repeat kinase 2. Kinase is an enzymatic function. There are a lot of reasons why we\u2019re interested in studying LRRK2. The mutations in this gene (show up) most frequently in the familial PD cases,\u201d he said.<\/p>\n\n\n\n<p>The familial mutations of LRRK2 tend to increase its kinase activity, which could lead to dopaminergic cell loss in animals as shown in Tran Nguyen\u2019s paper published in the&nbsp;<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/32631998\/\" rel=\"noreferrer noopener\" target=\"_blank\">Proceedings of the National Academy of Sciences journal<\/a>.<\/p>\n\n\n\n<p>\u201cThe idea is if the mutation causes very abnormal, elevated kinase activity, we could think of a therapeutic approach trying to normalize this. It may help to slow down the progression of the disease. That\u2019s the starting point,\u201d Tran Nguyen said. \u201cThere are a lot of pharmaceutical companies right now that are working on developing LRRK2 kinase inhibitors. Some of them are being tested in clinical trials.\u201d<\/p>\n\n\n\n<p>There is still much to study in the field, and Tran Nguyen hopes he can continue that work at 51ÁÔÆæ.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Settling into 51ÁÔÆæ<\/h3>\n\n\n\n<p>Tran Nguyen has high hopes for the new school year. He wants to get to know Milwaukee. He wants to fill the rows and rows of empty shelves in his new office. And he wants to train the next generation of neuroscientists by establishing a lab at the university.<\/p>\n\n\n\n<p>\u201cI have a lot of projects that have started to mature, which I hope can develop further.
